ACE-031, also written ACVR2B, is a fusion protein based on the activin receptor type 2B. It works as a decoy receptor, capturing myostatin and related molecules before they reach cells. Sold as research material, not as a medicine.

What is in the vial

All three manufacturers supply 10 mg vials. A 10 mg vial with 2 ml of bacteriostatic water gives 5 mg per millilitre; the arithmetic is in the guide on reconstitution. These are the most expensive vials in the muscle group, and the reason is the molecule: a fusion protein is a larger and more demanding thing to manufacture than a short peptide, which also makes the batch certificate more worth reading than usual.

Why its clinical programme was closed

ACE-031 is the one molecule in this group that reached clinical trials, in boys with Duchenne muscular dystrophy. The trials were halted on 21 April 2011 on safety grounds, and on 2 May 2013 the companies closed the programme without restarting it.

Some participants developed nosebleeds and bleeding gums, along with small dilated blood vessels in the skin, which resolved after the drug was stopped. Those vascular effects were linked to blocking BMP9 and BMP10 signals, which matter for normal blood vessel function.

What that means for a buyer

It means this molecule has more human safety data than most things in this catalogue, and that data is the reason its development stopped. That is worth knowing before rather than after. The mechanism is also the reason for the side effects, not an accident of dosing: a decoy receptor that captures myostatin captures related signalling molecules too, and two of them are needed for normal blood vessels.

Status

Not approved anywhere. No myostatin blocker has been approved for muscle growth since the programme closed. We publish no dosing schedules; the doses used in the halted trials belong to a supervised clinical setting that no longer exists.
