ACE-031, 10 mg. ACE-031, also written ACVR2B, is a fusion protein working as a decoy receptor: it captures myostatin and related molecules before they reach cells.

What is in this vial

All three brands of ACE-031 here come in the same 10 mg vial, so the choice between them is a choice of manufacturer and price, not of quantity. The powder has no concentration of its own: it is set when you add the water. This vial with 2 ml of bacteriostatic water gives 5 mg per millilitre, and with twice that volume, half as much.

The conversion of an amount into insulin syringe units is worked through in the guide on syringe units, and the rules that change once the powder is dissolved are in storage.

Bio-Peptide as the manufacturer

Bio-Peptide has the widest catalogue of the three brands we stock: it is the only one supplying glutathione, NAD+, GHK-Cu, MOTS-C, SLU-PP-332 and semaglutide, so an order from this manufacturer can usually be filled from one supplier. Every vial carries a code under the hologram and a batch certificate, and how to check both is described on the verification page.

What the evidence says

It is the one molecule in this group that reached clinical trials. Those trials were halted on safety grounds in 2011 and the programme closed in 2013. The full account, with the sources and the limits, is on the ACE-031 page, and the background in the guide on follistatin 344 and ACE-031.

We publish no dosing schedules. Sold as research material, not as a medicine.

ACE-031 is a fusion protein rather than a short peptide, which makes it one of the more demanding items in any catalogue to manufacture. Bio-Peptide fills it in the same 10 mg vial as the other two brands, so here the batch certificate is the document worth reading before the price.

Concentration in this vial

Water added Concentration Per unit on a U-100 syringe
1 ml 10 mg/ml 100 mcg
2 ml 5 mg/ml 50 mcg
5 ml 2 mg/ml 20 mcg

The table is arithmetic and nothing else: it converts the 10 mg in this vial into the concentration you get for a chosen volume of bacteriostatic water. It is not a dosing recommendation, and we publish none.

The same compound is sold here in two other versions: Genheal, 10 mg and Nouveaux Ltd., 10 mg. The molecule is identical in each; what differs is the manufacturer. All three hold the same amount, so the choice here is the manufacturer and its batch certificate, not the size.

Also from Bio-Peptide in this group

Within Muscle peptides, Bio-Peptide also supplies DES(1-3) IGF-1 1 mg, Follistatin 344 1 mg and IGF-1 LR3 1 mg.

What myostatin is and why block it

Myostatin, also labelled GDF-8, is a protein of the TGF-beta superfamily that restrains the growth of skeletal muscle. Animals and people with a mutation that switches off the myostatin gene have clearly more muscle mass. That observation is the basis of the whole idea of blocking this protein with a drug.

ACE-031 is a fusion protein based on the activin receptor type 2B joined to an antibody fragment. It works as a decoy receptor: it captures myostatin, activin A, activin B and related molecules before they reach muscle cells. That is where the abbreviation ACVR2B in the product name comes from.

The history of the clinical trials

ACE-031 was studied in boys with Duchenne muscular dystrophy, where blocking myostatin was meant to support muscle growth and repair. Acceleron Pharma and Shire halted those trials on 21 April 2011, and on 2 May 2013 they closed the whole programme without resuming it. The reason was safety: some participants had nose and gum bleeding and small dilated blood vessels in the skin, which resolved after stopping. They were linked to blocking BMP9 and BMP10 signals through the activin receptor, which matter for normal blood vessel function.

For a buyer this means the mechanism was tested in clinical conditions and showed both an effect on muscle and an unexpected side effect. We do not write that ACE-031 builds muscle in a proven and safe way: for healthy people there are no clinical data confirming it.

Two concentrations in this box

The Bio-Peptide box holds two solvent ampoules. Dissolving the whole 10 mg in one millilitre gives 100 mcg per syringe unit; in two millilitres it gives 50 mcg per unit and a scale twice as fine. The choice is made once, when dissolving.

Who this vial is for

You are interested in the myostatin pathway and want a choice of concentration: this is the vial. You are looking for the second molecule aimed at the same pathway: that is follistatin 344.

A study in healthy people: what one injection did

ACE-031 is one of the few myostatin blockers that increased muscle mass in healthy people. The study was published in 2013 in Muscle and Nerve. It was a double-blind, placebo-controlled trial in 48 healthy postmenopausal women. Each received one subcutaneous injection of ACE-031 or placebo.

What was measured Result at the highest dose, day 29
Whole-body lean mass plus 3.3 percent
Thigh muscle volume on MRI plus 5.1 percent
Half-life 10 to 15 days
Tolerability generally good, redness at the injection site

Both muscle changes were statistically significant, and they came from a single injection. The blood level of ACE-031 rose in proportion to the dose. Blood markers also suggested a favourable effect on bone and fat tissue turnover.

What the study in boys with dystrophy showed

ACE-031 was also tested in boys with Duchenne muscular dystrophy, in a randomised, placebo-controlled study published in 2017. The boys received injections every 2 to 4 weeks. The treated groups showed a trend towards keeping their distance in the 6-minute walk test, while in the placebo group the distance fell.

There were also trends towards higher lean mass and bone density and lower fat mass. The study was stopped after the second dosing schedule because of nosebleeds and dilated small blood vessels, although there were no serious side effects. The authors judged myostatin blocking a promising direction for treating this disease. All the results are gathered in the guide Follistatin 344 and ACE-031: myostatin blockers.

Questions

How much ACE-031 is in this vial?

10 mg of lyophilised powder from Bio-Peptide, in one vial.

What concentration does 2 ml of water give?

5 mg per millilitre, which is 50 mcg per unit on a U-100 insulin syringe. Other volumes are in the table above.

Is this the same molecule as the Genheal vial?

Yes. ACE-031 is the same compound in every vial here, and both hold 10 mg. What differs is the manufacturer and the batch certificate that comes with it.
